Oxidative Stress, Chromium-Resistance and Uptake by Fungi: Isolated from Industrial Wastewater BABT
Elahi,Amina; Rehman,Abdul.
ABSTRACT Trichosporon asahii and Rhodotorula mucilaginosa isolated from wastewater effluents were identified as chromium-resistant yeasts. Cr(VI) concentrations at 8 mM and 6 mM were inhibitory for R. mucilaginosa and T. asahii. Remarkably elevated GSH (69.88 ± 10.01) and GSSG (11.24 ± 0.96) was observed under metal stress in T. asahii as compared to R. mucilaginosa GSH (18.95 ± 3.19) and GSSG (3.7 ± 2.74) mM g-1 8 level. Statistical analysis revealed significantly higher GSH/GSSG ratio in both strains. NPSH (29.84 ± 0.54) level in T. asahii was much higher than in R. mucilaginosa (6.05 ± 0.24). Chromate reductase (ChR) was assayed and its activity was optimum at 50°C (pH 6) in T. asahii while R. mucilaginosa showed higher activity at 30°C (pH 7). Activity...

Paralytic phycotoxin uptake by scallops (Pecten maximus) ArchiMer
Lassus, Patrick; Bardouil, Michele; Ledoux, Martial; Murail, Ingrid; Bohec, Madeleine; Truquet, Philippe; Frémy, Jean-marc; Rohmer, Véronique.
It is difficult to apply sanitary standards for mollusc contamination by paralytic shellfish poisoning to animals not consumed whole. This problem is illustrated by the 1990 embargo on a Japanese shipment of frozen scallop muscle and gonad: the sanitary threshold applied to digestive gland (in Japan) does not guarantee that muscle and especially gonad are toxin-free. Accordingly, we performed experimental contaminations of scallops (Pecten maximus) from Port-en-Bessin (Normandy, France) using a toxic Japanese strain of Alexandrium tamarense. During the contamination/decontamination experiment on different tissues (digestive gland, muscle, gonad), extracts were obtained using cold acetic acid 0.1 N to maintain the perfect integrity of the toxin profile....

Past and present platinum contamination of a major European fluvial-estuarine system: Insights from river sediments and estuarine oysters ArchiMer
Abdou, M.; Schaefer, J.; Cobelo-garcia, A.; Neira, P.; Petit, J. C. J.; Auger, Dominique; Chiffoleau, Jean-francois; Blanc, Gerard.
Platinum Group Metals (PGM) are modern, technologically relevant elements for which (i) the anthropogenic cycle has outcompeted the natural cycles and (ii) environmental behavior, fate and impact are still widely unknown. Stripping voltammetry was used for accurate determinations of platinum (Pt) in historical records of river sediments and estuarine oysters from the Gironde fluvial-estuarine continuum (SW France) comprising the Lot River. Sediment cores from the Lot River, dated from 1952 to 2001, showed past Pt contamination due to former industrial (smelter) activities in the Lot River watershed. These samples revealed the phasing-out of a historical Pt contamination with Pt/Th (Thorium) values of 11 × 10− 5 ± 0.79 × 10− 5 for the deepest part of the...

PCB concentrations in plankton size classes, a temporal study in Marseille Bay, Western Mediterranean Sea ArchiMer
Tiano, Marion; Tronczynski, Jacek; Harmelin-vivien, Mireille; Tixier, Celine; Carlotti, Francois.
PCB levels in plankton were investigated in the Bay of Marseille, Western Mediterranean Sea, between September 2010 and October 2011. Concentrations of PCB congeners (CB 18, CB 52, CB 101, CB 118, CB 138, CB 153, CB 180) were determined in three plankton size-classes (60–200, 200–500 and 500–1000 μm) together with different parameters: chlorophyll content, plankton dry-weight biomass, carbon and nitrogen stable isotope ratios and plankton-community structure. The ∑PCB7 concentrations ranged between 14.2 and 88.1 ng g−1 d.w., for all size classes and all sampling periods. The results do not show the biomass dilution effect and indicate moderate but significant biomagnification with plankton trophic position estimated by δ15N signatures. Equilibrium with...

Po-210 and Pb-210 in the tissues of the deep-sea hydrothermal vent mussel Bathymodiolus azoricus from the Menez Gwen field (Mid-Atlantic Ridge) ArchiMer
Charmasson, Sabine; Le Faouder, Antoine; Loyen, Jeanne; Cosson, Richard P.; Sarradin, Pierre-marie.
The hydrothermal deep-sea vent fauna is naturally exposed to a highly specific environment enriched in potentially toxic species such as sulfides, metals and natural radionuclides due to the convective seawater circulation inside the oceanic crust and its interaction with basaltic or ultramafic host rocks. However, data on radionuclides in biota from such environment are very limited. An investigation was carried out on tissue partitioning of Po-210 and Pb-210, two natural radionuclides within the U-238 decay chain, in Bathymodiolus azoricus specimens from the Mid-Atlantic Ridge (Menez Gwen field). These two elements showed different distributions with high Pb-210 levels in gills and high Po-210 levels in both gills and especially in the remaining parts of...

Selenium biofortification on garlic growth and other nutrients accumulation Horticultura Brasileira
Pérez,María B; Lipinski,Víctor Mario; Filippini,María Flavia; Chacón-Madrid,Katherine; Arruda,Marco Aurelio Z; Wuilloud,Rodolfo G.
ABSTRACT Selenium (Se) is an essential element for humans and has anti-cancer function. Garlic can accumulate Se, so it is an option to Se supplementation in the human diet. The aim of this research was to study Se uptake and accumulation during garlic growth. Four doses of Na2SeO4 and Na2SeO3 solution were applied in the substrate (0, 5, 10 and 15 kg ha-1 Se) for one time in August 2014, with a random plot design and 3 replicates on garlic clone Rubi INTA. Three harvests were made, in September, October and December 2014. After each harvest, leaves, bulbs and roots were separated and conditioned (peeled and chopped), lyophilized, and finally acid-digested prior to Se, Mg, Zn, Mn, Cu, Fe, P and S determination by inductively coupled plasma mass...

Spatial variation of metal bioaccumulation in the hydrothermal vent mussel Bathymodiolus azoricus ArchiMer
Cosson, R; Thiebaut, E; Company, R; Castrec Rouelle, M; Colaco, A; Martins, I; Sarradin, Pierre-marie; Bebianno, M.
The variability of the bioaccumulation of metals (Ag, Cd, Cu, Fe, Mn and Zn) was extensively studied in the mussel Bathymodiolus azoricus from five hydrothermal vent sites inside three main vent fields of increasing depth along the Mid-Atlantic Ridge: Menez Gwen, Lucky Strike and Rainbow. Metal bioaccumulation varied greatly between vent fields and even between sites inside a vent field with B. azoricus showing a great capacity to accumulate metals. The bioaccumulation of these metals also varied significantly among tissues. The main target was the gills where metals were mainly associated with soluble compounds whereas in the digestive gland they were mainly associated with insoluble compounds. Storage of metals under insoluble forms in B. azoricus seems...

Total and organic Hg concentrations in cephalopods from the North Eastern Atlantic waters: Influence of geographical origin and feeding ecology ArchiMer
Bustamante, Paco; Lahaye, V; Durnez, C; Churlaud, C; Caurant, Florence.
Total (T-Hg) and organic (O-Hg) mercury concentrations and tissue distribution were examined in 20 species of cephalopods (n = 278) from the north eastern Atlantic waters, i.e. from the Bay of Biscay to the Faroe Islands. Concentrations of T-Hg in whole cephalopods showed elevated variations among species, i.e. from 40 to 3560ng g(-1) dwt, but a low variability within each species (mean CV%=39%). With the exception of oceanic squids, the digestive gland globally displayed higher T-Hg concentrations than the remaining tissues. In contrast, O-Hg concentrations determined in selected species were generally higher in the remaining tissues. Despite higher T-Hg concentrations, the digestive gland weakly contributed to the total body burden of both T-Hg and O-Hg...

ABSTRACT This is the first study to investigate mercury (Hg) contamination in felid species in the Brazilian Amazon. We collected 26 fur samples from wild felids of four species (Puma concolor, Panthera onca, Leopardus pardalis and Leopardus wiedii) occurring in the Mamirauá and Amanã sustainable development reserves, in the state of Amazonas. Samples were from museum specimens, except for five P. onca samples collected from free-living individuals. Total Hg concentrations ranged from 0.12 to 48.1 µg g-1. Concentrations of Hg did not differ significantly between museum specimens and live individuals of P. onca, but varied significantly among species, with significantly higher concentrations for P. onca and L. pardalis, which could be related to factors...

From 1966 to 2015, the Gardanne alumina refinery discharged some 20 million tons of bauxite residue (called red mud) into the Cassidaigne Canyon (northwest French Mediterranean) with impacts on local ecosystem functioning. Although these red muds contained high levels of trace elements (TE), in particular titanium (Ti), vanadium (V), aluminum (Al) and arsenic (As), surprisingly, their impacts on fish contamination levels and the risk related to fish consumption have been little studied until now. Here, 11 trace elements (Al, As, Cd, Cr, Co, Cr, Mn, Ni, Pb, Ti and V) were analyzed in muscle and, when possible, liver, from 1308 fish of 26 species from an impacted zone in the vicinity of the Cassidaigne Canyon and a reference zone, unaffected by red mud...

Trace metal concentrations in the muscle of seven marine species: Comparison between the Gulf of Lions (North-West Mediterranean Sea) and the Bay of Biscay (North-East Atlantic Ocean) ArchiMer
Mille, Tiphaine; Cresson, Pierre; Chouvelon, Tiphaine; Bustamante, Paco; Brach-papa, Christophe; Bruzac, Sandrine; Rozuel, Emmanuelle; Bouchoucha, Marc.
Concentrations of 6 trace metals (Ag, Cd, Cu, Ni, Pb, Zn) in the muscle of 2 sharks (Galeus melastomus and Scyliorhinus canicula), 4 teleosts (Helicolenus dactylopterus, Lepidorhombus boscii, Micromesistius poutassou and Phycis blennoides) and 1 crustacean (Nephrops norvegicus) were compared between the Bay of Biscay (Atlantic Ocean) and the Gulf of Lions (Mediterranean Sea). Although average concentrations and the trace element pollution index were generally higher in the Gulf of Lions, significant differences between the two ecosystems were only found for Zn for Helicolenus dactylopterus, and for Ag and Cu for the crustacean N. norvegicus. Moreover, some relationships between trophic level or size and metal concentrations were found for these two...

Uptake of free and complexed silver ions by different strains of Rhodotorula mucilaginosa BJM
Gomes,Newton C.M.; Rosa,Carlos A.; Pimentel,Patrícia F.; Mendonça-Hagler,Leda C.S..
Five strains of Rhodotorula mucilaginosa were tested for the ability to accumulate free and complexed silver ions by metabolism-dependent and -independent processes. The ability to take up Ag+ was observed in both live and dead biomass, whereas silver dicyanide [Ag (CN)2-] uptake was strictly glucose dependent. In contrast to Ag (CN)2-, glucose addition inhibited by 16 to 25% the Ag+ uptake rate of living UFMG - Y02, Y27, and Y35 cells, while strains CBS 316 and UFMG-Y01 showed an improved uptake rate of about 115% and 13%, respectively. The Langmuir sorption model was used to evaluate the silver sorption capability of the R. mucilaginosa strains. The calculated q max value suggested that R. mucilaginosa strains UFMG-Y27 had the highest loading capacity....

Use of rotavirus virus-like particles as surrogates to evaluate virus persistence in shellfish ArchiMer
Loisy, Fabienne; Atmar, R; Le Saux, Jean-claude; Cohen, J; Caprais, Marie-paule; Pommepuy, Monique; Le Guyader, Soizick.
Rotavirus virus-like particles (VLPs) and MS2 bacteriophages were bioaccumulated in bivalve mollusks to evaluate viral persistence in shellfish during depuration and relaying under natural conditions. Using this nonpathogenic surrogate virus, we were able to demonstrate that about 1 log(10) of VLPs was depurated after 1 week in warm seawater (22 degrees C). Phage MS2 was depurated more rapidly (about 2 log(10) in 1 week) than were VLPs, as determined using a single-compartment model and linear regression analysis. After being relayed in the estuary under the influence of the tides, VLPs were detected in oysters for up to 82 days following seeding with high levels of VLPs (concentration range between 10(10) and 10(9) particles per g of pancreatic tissue)...

Variability of PCB burden in 5 fish and sharks species of the French Mediterranean continental slope ArchiMer
Cresson, Pierre; Fabri, Marie-claire; Miralles, Francoise Marco; Dufour, Jean-louis; Elleboode, Romain; Sevin, Karine; Mahe, Kelig; Bouchoucha, Marc.
Despite being generally located far from contamination sources, deep marine ecosystems are impacted by chemicals like PCB. The PCB contamination in five fish and shark species collected in the continental slope of the Gulf of Lions (NW Mediterranean Sea) was measured, with a special focus on intra- and interspecific variability and on the driving factors. Significant differences occurred between species. Higher values were measured in Scyliorhinus canicula, Galeus melastomus and Helicolenus dactylopterus and lower values in Phycis blennoides and Lepidorhombus boscii. These differences might be explained by specific abilities to accumulate and eliminate contaminant, mostly through cytochrome P450 pathway. Interindividual variation was also high and no...

Variation of heavy metal concentrations (Ag, Cd, Co, Cu, Fe, Pb, V, and Zn) during the life cycle of the common cuttlefish Sepia officinalis ArchiMer
Miramand, Pierre; Bustamante, Paco; Bentley, Daniel; Koueta, Noussithe.
The developmental changes in the concentration of 8 essential and non-essential heavy metals (Ag, Cd, Cu, Co, Fe, Pb, V, Zn) in the tissues (digestive gland, cuttlebone and whole animal) of the common cuttlefish Sepia officinalis collected in the bay of the river Seine were monitored from the end of the embryogenesis until the adult reproductive stage. Compared to embryos, juveniles after hatching displayed much higher concentrations of Ag, Cu, Fe and Zn, suggesting an efficient incorporation from seawater. Conversely, the amounts of Cd, Pb and V in hatchlings remained constant suggesting that these metals are barely bioavailable for juveniles. Once the juveniles start to feed, the digestive gland appears to play a major role in the storage of all metals....
